Preliminary work to test general Ruggedness of the Assay
Capture step at maximum and minimum time.
(see Figure 2 below).
It was found during assay development that there is no meaningful difference between short (15hr) or long (24hr) capture incubation times; therefore no additional validation testing related to capture antibody incubation time was performed as part of this validation study.

Extreme assay conditions were tested to see the effect on RP value of a vaccine.
ELISA testing with sample, conjugate, and substrate steps tested at maximum incubation times (3hr 5min, 1hr 5min, and 32min, respectively) vs minimum incubation times (2hr 55min, 55min, and 28min, respectively) were tested (see Table 1 below).
It was concluded that there was little difference between incubation at standard time vs minimum time and that the RP at maximum incubation time was 0.3RP lower (1.4) than the standard time RP (1.7).
These preliminary data were only from one session and further investigation were done during assay verification ruggedness testing.
