These colonies appear as a translucent lawn of growth covering the surface of the plate. The phages form clear patches in the bacterial lawn, called plaques. The plaques form because the phages have bust open the bacteria at hat site on the plate. By counting the number of plaques, and multiplying by the serial dilution factor, one can determine the titer, which is the number of phage particles in the original phage culture.
