Preparation of the Prevegetative Cultures
Spore suspension from the solid surface seed culture prepared with sterilized water is used to inoculate sterilized malt extract media in Erlenmeyer flasks.
The cultures are incubated on a rotatory shaker for 46-50 hours at 25-27oC.
Sterilized malt extract media in Erlenmeyer flasks are inoculated with 10% (v/v) prevegetative culture.
The fungal cultures are incubated for 44-52 hours on a rotary shaker at 25-27°C and **pressed to a sterilized inoculating can** under aseptic conditions.
