A minha dúvida aqui é se em PTBR colocamos 2:20 apenas ou 2:20s quando se trata de video, pois na verdade seriam 2min20s.

Na verdade, na tradição gráfica do português não são usadas aspas retas. O nome do sinal é plica: simples (′) ou dupla (″). Elas são usadas para simbolizar:
