En 1854, James Clerk Maxwell décrivait la lentille optique parfaite : sphérique, permettant d'éliminer toute forme d'aberration, et ainsi de donner des images d'une netteté absolue, elle nécessite néanmoins une variation continue de l'indice de réfraction du matériau, du centre jusqu'à la périphérie. Les cristallins des yeux des calmars présentent justement cette propriété, permettant à l'animal d'obtenir, dans les sombres profondeurs sous-marines, une image parfaitement claire de son environnement. En analysant la composition en protéines du cristallin de l'animal, Jing Cai, de l'université de Pennsylvanie, et ses collègues ont levé le voile sur ce biomatériau remarquable.
Le cristallin est en effet un gel constitué principalement d’eau, et d’un mélange de protéines globulaires, appelées S-cristallines. C’est la concentration hétérogène de ces dernières qui est responsable de la variation de l’indice de réfraction. Au centre, Les chercheurs ont trouvé que les S-cristallines formaient un réseau très dense, avec très peu d’eau, alors qu’en périphérie, elles ne représentent que 4 % du matériau. Pourtant, cette répartition inégale restait difficile a expliquer, compte tenu de la propension des protéines à diffuser librement dans ce gel.
En étudiant les ARN messagers codant pour ces protéines, Jing Cai et ses collègues ont identifié non pas une, mais 53 S-cristallines différentes, réparties inégalement entre le centre et la périphérie du cristallin : ne différant les unes des autres que par environ 15 % de leurs acides aminés, elles présentent toutes la même structure centrale, mais portent à leur surface des boucles d’acides aminés désordonnées et de longueurs très variables. Plus la distance au centre du cristallin est grande, plus la proportion de protéines aux boucles courtes diminue. De 34 % de la masse de protéine totale au centre, elles ne représentent plus que 5 % à la périphérie, remplacées graduellement par des protéines aux boucles de plus en plus longues.
« Les auteurs ont montré que ces boucles désordonnées permettent de limiter les interactions entre protéines. Plus elles sont longues, moins les protéines s’agrègent les unes aux autres » explique Sophie Sacquin-Mora, chargée de recherche CNRS à l’Institut de biologie physico-chimique, à Paris.
Ainsi, les simulations numériques ont établi qu’au centre, les S-cristallines aux boucles les plus courtes sont chacune liées à six voisines, formant ainsi un réseau dense, sans pour autant précipiter, ce qui provoquerait inévitablement une opacification du cristallin. D’après les calculs, en périphérie, les protéines aux boucles longues ont seulement deux voisines, conduisant à un réseau beaucoup moins dense, et donc un gel beaucoup riche en eau, et ainsi un indice de réfraction plus faible.
Les auteurs ont également étudié le processus de formation du cristallin. « La synthèse de ces S-cristallines nécessite leur diffusion loin des ribosomes qui les fabriquent. Lorsque le milieu est peu dense, toutes les protéines peuvent diffuser, et sont donc synthétisées. Lorsqu’il l’est davantage, les protéines à grandes boucles ne peuvent plus diffuser; leur synthèse stoppe, au profit des plus petites », détaille Sophie Sacquin-Mora, reliant ainsi la variation de composition du biomatériau avec sa densité en S-cristallines.

In 1854, James Clerk Maxwell described the perfect lens: spherical, it would permit the elimination of any optical aberration and so produce images of absolute clarity. However, it would also require the refractive index of the material from which it was made to vary continuously between the centre and the edge.
The lenses in squids' eyes exhibit precisely this property, allowing them to obtain a perfectly clear image of their surroundings in the dark undersea depths. By analysing the protein composition of the lenses in the animals' eyes, Jing Cai of the University of Pennsylvania and her colleagues have lifted the veil on this remarkable biomaterial.
The lens is effectively a gel composed mainly of water along with a blend of globular proteins called S-crystallins. It is the different concentrations of S-crystallins in each part of the lens that determine the refractive index at that point.
The researchers found that at the centre of the lens, the S-crystallins formed a very dense network, with very little free water, while at its edges they made up only 4% of the material. However, this uneven distribution remained difficult to explain, given the ease with which proteins diffuse through the gel.
By studying the messenger RNA that codes for these proteins, Jing Cai and her colleagues have identified not one, but 53 different S-crystallins, present in varying proportions from the centre to the edge of the lens.
These proteins only differ from each other by around 15% of their amino acids. Each has the same central structure, but the disorganised loops of amino acids that project from its surface vary considerably in length. The gel contains fewer proteins with short loops the further it is from the centre of the lens. Forms with short loops make up 34% of the protein by mass at the centre, but no more than 5% at the edge, having gradually been replaced by forms with ever longer loops.
“The authors have shown that these disorganised loops can limit interaction between proteins. The longer they are, the less the proteins link to each other” explains Sophie Sacquin-Mora, CNRS research associate at the Institute of Physico-Chemical Biology in Paris.
Computer simulations have shown that at the centre of the lens, the S-crystallins with the shortest loops are each linked to six neighbours. They form a dense network without sticking together into solid clumps, which would inevitably cloud the lens. The results show that at the edges of the lens, the proteins with long loops have only two neighbours, forming a much less dense network. This leads to a water-rich gel with a lower refractive index.
The authors have also studied the process by which the lens is formed. "The synthesis of these S-crystallins requires them to diffuse a long way from the ribosomes that make them. In a low density medium, all of the proteins are able to diffuse, and are therefore synthesised. When it is denser, the proteins with longer loops can no longer diffuse; their synthesis is halted in favour of the smaller ones", explains Sophie Sacquin-Mora, linking the variation in composition of the biomaterial to its density in S-crystallins.

1. UNE EXONÉRATION SOCIALE PAR JEI QUASIMENT À SON NIVEAU D’AVANT LA RÉFORME DE 2011
Sur la période 2004-2010, le montant des exonerations de cotisations sociales accordées aux JEI a augmenté progressivement et a plus que doublé, en lien avec l’accroissement naturel du nombre de bénéficiaires dans le temps. Il est passé de 62 M€ en 2004 à 144 M€ au cours de 2010 (graphique 1). Sur cette période, chaque JEI a perçu, en moyenne, 50 k€ d’exonération de cotisations sociales par année, ce qui correspond approximativement à la prise en charge par l’État du salaire d’un chercheur par JEI.
En 2011, ces exonérations reculent de 37 % en raison de leur plafonnement et d’une dégressivité dans le temps, tous deux instaurés par la loi de finances. Le montant d’exonération par JEI s’établit alors à 30 k€ par an en moyenne. L’assouplissement de cette réforme en loi de finances rectificative 2011 entraîne une hausse du montant d’exonération par JEI à 35 k€ par an en moyenne en 2012 et 2013.
La dernière réforme du dispositif, en loi de finances 2014, supprime la dégressivité des exonérations sociales, tout en maintenant un plafond, mais en le relevant, et étend le dispositif à des dépenses d’innovation (MINEFI, 2013). Elle produit ses premiers effets dès 2014 : les montants des exonérations de cotisations sociales accordés, par JEI et au total, sont depuis en progression. Le montant total s’élève à environ 170 M€ en 2016 et le montant moyen atteint 47 k€, en hausse de 5 % par rapport à 2015, même s’il demeure légèrement en deçà de son niveau de l'année 2010. La loi de finances pour 2017 a prorogé le dispositive sur la période 2017-2019 (MEFI, 2016).

3. UNE CONCENTRATION DANS LES SECTEURS INFORMATION ET COMMUNICATION, ET ACTIVITÉS SCIENTIFIQUES ET TECHNIQUES
Depuis la création du dispositif, la répartition sectorielle des JEI est stable. En 2016, 85 % relèvent du secteur Information et communication (44 %) et des Activités spécialisées, scientifiques et techniques (41 % ; tableau 2 page suivante). À titre de comparaison, un peu plus de la moitié des PME hors micro-entrepreneurs et réalisant des activités de RDI appartiennent à ces mêmes secteurs. Les JEI de ces deux secteurs concentrent 87 % des salariés et 88 % du montant des exonérations de cotisations sociales octroyées, soit 150 M€ au total.
À l’inverse, l’Industrie manufacturière, qui regroupe un quart des PME hors micro-entrepreneurs réalisant des activités de RDI, ne totalise que 7 % des JEI. Ces dernières bénéficient d’environ 6 % des exonérations sociales.

1. SOCIAL SECURITY EXEMPTIONS PER JEI CLOSE TO PRE-2011 REFORM LEVELS
Over the period 2004-2010, the volume of exemptions from social security contributions that were granted to JEIs progressively increased—more than doubling—in line with the natural increase in the number of eligible companies over time. It went from €62M (£42M) in 2004 to €144M (£124M) during 2010 (Figure 1). Over this period, each JEI received on average €50,000 (£37,500) of social security contribution exemptions per annum, which is broadly equivalent to the State paying the salary of one researcher for each JEI.
The 2011 French loi de finances (finance bill) capped exemptions and made them degressive over time for each SME, the two measures together causing total exemptions to fall by 37%. Consequently, exemptions per JEI averaged €30,000 (£26,000) per annum. The relaxation of this reform by the same year’s loi de finances rectificative (amending finance bill) led to an increase in the average amount of exemptions per JEI to €35,000 (£29,000) per annum in 2012 and 2013.
The most recent amendment to the scheme, in the 2014 loi de finances, ended degression of social security exemptions, while retaining an increased cap, and extended the scheme to cover expenditure on innovation (MINEFI, 2013). These changes began to take effect from 2014 and the total value of exemptions granted from social security contributions, both per JEI and in total, has been rising ever since. The total amount reached around €170M (£139M) in 2016 and the average amount reached €47,000 (£39,000), up 5% compared to 2015, although it remains slightly below its 2010 level in Euros. The 2017 loi de finances extended the scheme to cover the period 2017-2019 (MEFI, 2016).

2. CONCENTRATION IN THE ICT AND SCIENCE & TECHNOLOGY SECTORS
Since the creation of the scheme, the distribution of JEIs across market sectors has been stable. In 2016, 85% were in either the Information Communication Technology (ICT) (44%) or the Science & Technology (41%) sectors (table 2 on following page). By way of comparison, if micro-entrepreneurs (a light-touch taxation category for self-employed workers) are excluded then just over half of SMEs that carry out RDI belong to these two sectors. The JEIs from these sectors account for 87% of the employees and 88% of the total social security contribution exemptions granted, €150M (£123M) in total.
Conversely, manufacturing industry, again excluding micro-entrepreneurs, makes up a quarter of SMEs carrying out RDI but represents only 7% of JEIs. This group benefits from about 6% of the total social security exemptions.

My first Masters is in Aeronautics and Astronautics, which included a final year studying in French at the École Supérieure de Technique Aéronautique et de Construction Automobile (ESTACA). I more recently gained a second Masters Degree, this time in translation.
